ABSTRACT

AIM: To study the protective effect of fenofibrate on diabetic retinal neurodegeneration and observe its effect on miR-26a-5p and its target gene PTEN in the retinal of diabetic mice.METHODS: Diabetic mice models were established and they were gavaged by fenofibrate. H& E staining and transmission electron microscopy were used to observe the impairments of retinal neurons. Real-time PCR was used to examine the expression of miR-26a-5p, and Western blotting was employed to measure the expression of phosphatase and tensin homologue(PTEN)in the retina of diabetic mice. The expression level of nuclear factor-κB(NF-κB), interleukin-1β(IL-1β)and the morphology of neural tissues were observed.RESULTS: When compared with the diabetic mice, fenofibrate significantly attenuated the damage to retinal ganglion cells and the atrophy of retinal nerve fiber layer. While the level of miR-26a-5p was increased and the levels of PTEN and inflammatory mediators were significantly decreased in the retina of fenofibrate treated diabetic mice, with significant statistical significance(P<0.05).CONCLUSIONS: Fenofibrate protects against diabetic retinal neurodegeneration by upregulating miR-26a-5p and inhibiting PTEN, attenuating the inflammatory response and alleviating retinal cell injury.

ABSTRACT

Objective@#To explore the interrelationships among fathers’ parenting effectiveness, urban children’s sociality and the acceptance of “second-born children” by children (including only children and “first-born children”), and to provide a reference for interventional measured for improving children’s acceptance rate of "second-bron children".@*Methods@#Stratified cluster sampling method was used to select 3 to 9-year-old 778 only children and “first-born children” in two kindergartens and grades 1-3 of two primary schools in a city, and the children and their fathers were investigated with questionnaires survey regarding general information of both fathers and children, the acceptance degree of “second-born children”, the sociality of children and the parenting effectiveness of fathers.@*Results@#Children in different grades had different acceptance levels of “second-born children” and the difference was statistically significant (F=2.89,P<0.05). Correlation analysis showed that the acceptance of “second-born children” was negatively correlated with the interaction between the only children and “first-born children”, and positively correlated with the prosocial behavior of the only children and “first-born children”, both of which are statistically significant (r=-0.10,0.10,P<0.01). Father’s role efficacy was negatively correlated with onlychildren and “first-born-children” peer interaction problems(r=-0.15), and positively correlated with prosocial behavior(r=0.32), both of which were statistically significant (P<0.01). The structural equation model of father’s parenting effectiveness, children’s sociality and acceptance of “second-born children” showed that low father’s parenting effectiveness could positively predict children’s peer communication problems and negatively predict children’s prosocial behavior, and thus children’s peer communication problems could negatively predict the acceptance degree of only children and “first-born children” to “second-born children”, and prosocial behaviors could positively predict the acceptance degree of “second-born children”.@*Conclusion@#Promoting father’s parenting efficiency can facilitate the social development of children and the acceptance of siblings. It’s recommended that fathers actively participate in specific parenting matters for the only child and "one child",and consciously improve parenting effectiveness.

ABSTRACT

Purpose To explore the value of Doppler ultrasound in the diagnosis of congenital hypertrophic pyloric stenosis (CHPS) by observing pylorus. Materials and Methods Sixyt-five children with CHPS diagnosed by ultrasound in Neonatology Department of Guangzhou First People's Hospital from 2006 to 2015 were retrospectively analyzed. CHPS group and 50 infants in control group were examined by Doppler ultrasound to observe the features of blood flow distribution and blood flow grading, and Doppler spectrum analysis was performed to observe contrast agent passing through pyloric canal. Results The difference of the thickness of muscular layer and mucous layer, and that of the long diameter and inner diameter of pyloric canal in CHPS group and control group all suggested marked statistical significance (P<0.01); the blood flow grading in muscular layer and mucous layer between the two groups showed statistical significance (t=13.33 and 18.77, all P<0.01). The blood flow velocity in muscular layer of CHPS group was (16.96±0.91) cm/s, resistance index (RI) 0.68±0.33, inner diameter of pyloric canal (1.98±0.33) mm. Conclusion Doppler ultrasound enables us to grasp the distribution and grading of blood flow of pylorus as well as the condition of contrast agent passing through pyloric canal, in which way the degree of pyloric stenosis can be evaluated by combining indexes such as Vmax and RI, and an objective basis for clinical choice of treatment can be provided, possessing high application value.
